IFNB1 is a cytokine signalling protein that belongs to the interferon family (IFN), which acts as part of the innate immune response. IFNB1 is a type I IFN and normally exerts antiproliferative, antiangiogenic, proapoptotic and immunemodulatory functions [4330].
IFNB1 has been shown to be part of the DNA damage response, a process involved in human ageing [4331]. In Terc knockout mice, which normally show accelerated ageing, inactivation of IFNB1 results in the abrogation of progeric phenotypes and extends lifespan. In human and mouse cells the accumulation of DNA damage results in the production of IFNB1 and triggers IFN signalling both in vitro and in vivo [4331]. In addition, inhibition of IFNB1 leads to decreased cell senescence, demonstrating that IFNB1 is a mediator of cell senescence [4331]. Thus, IFN signalling may act as a mechanism that links DNA damage to senescence and premature ageing.
