Depletion of sug using RNA interference reduced lifespan in flies and reduced the lifespan extending effects of dietary restriction by 37%. Sug was found to act downstream of Ire1, facilitating the metabolic shift towards lipid usage under dietary restriction. Sug expression rescued the short lifespan of Ire1 depleted flies on DR.
