GHRH is secreted in the hypothalamus to stimulate the synthesis and secretion of growth hormone (GH1). Defects in the human GHRH gene cause dwarfism while hypersecretion causes gigantism, but it is unknown whether the ageing process is affected in these patients. Declines in GH1 with age may be due to a deficiency in GHRH [1354]. Knockout mice live 46% longer than controls: Females live 43% longer and males 51% longer, though maximum lifespan was only increased (by 33%) in females. Both males and females and markedly smaller than controls [3280]. Given these results, it is possible that GHRH is involved in some aspects of human ageing.
