Mitochondrial uncoupling proteins create proton leaks across the inner mitochondrial membrane to uncouple oxidative phosphorylation from ATP synthesis resulting in energy being dissipated as heat. Mice with skeletal muscle-specific UCP1 expression had increased median survival, decreased adiposity, increased temperature and metabolic rate. The same genetic manipulation in APOE null mice decreased diet-induced atherosclerosis and in agouti yellow mice, a model of obesity, diabetes and hypertension were reversed [1850].
